Batch acclimatisation experiments were completed for the final construction of the advanced composite bioaugmentation inocula. The nexts steps included, as part of the project, testing and designing different variations of the DNA extraction protocol using the DNeasy PowerSoil Pro Kit. We experimented with different durations and intensities of mechanical lysis and evaluated the results both qualitatively (through agarose gel electrophoresis) and quantitatively (using Qubit measurements). We concluded that the method involving two cycles of 20 seconds each provided the best DNA yield and quality, which will be used in the next phases of the project. The implementation of lyophilisation with the developed optimum lyophilisation method for the consortia was also performed, and further results are pending.
